Well, it couldn't hurt. The locking nut provides tuning stability. Hell, even when unblocked, you could leave it unlocked. You'd be an idiot to do it, but you could.
Likewise with it blocked. When any guitar company puts a locking nut on a guitar, they probably don't throw the world's greatest tuners behind it. After all, the tuners really aren't meant to withstand abuse (that's what the nut's for). If I were you, I'd lock it. It provides much greater stability, and takes 10 seconds to do... why not?
