You need to stretch your hand out before you play.

If you give yourself a good warmup and you're still cramping up, re-examining your picking technique sounds like a good idea.
Make sure you yourself aren't forcing the tension into your right hand as you play. Watch for when you lock your wrist or raise your shoulders.
Practicing the riff with a metronome at a slow tempo and focusing your attention on keeping your entire body relaxed will help you realize what you're doing wrong.

also, once it starts to hurt, you might want to stop playing guitar for a week or so, cuz you can damage your hand/arms pretty bad from overdoing it to much. i got it a year ago or so, couldnt play guitar for like 2/3 months.
and yet, stretchign and warming up also might help... also, try to play it as relaxed as possible, once you feel your getting cramped up try to find a way to not cramp your muscles, it might save you alot of trouble lateron.

... and never underestimate the use of simple chord strumming when you're trying to advance your skills. Both as a warm-up and a bit of a break from the difficult stuff; I often do that when I feel that I'm beginning to tense up.

Practice it more slowly. Slow it down enough that you don't tense up. You should always be relaxed when playnig quickly. Don't increase your speed when practicing unless you can play with both hands relaxed. Tension is the biggest enemy of speed.
